The spade connectors on the speaker cable on my 210 sixty-five aren't marked to indicate + or -

Does it make a difference to which way it is connected as long as the wires between the speakers are ok relative to each other?

You can't really blow anything up, but, you would really like to have them in phase with one another (positive to positive and negative to negative). If they get out of phase then you will experience cancellation between the speakers which will give you a muddy-middy sound. One speaker will move out while the other speaker moves in during a half cycle. It shouldn't matter if you get the leads from the amp swapped as long as the speakers are wired for the same polarity.
